ADHP / Amplex Red Reagent


Although chromogenic peroxidase substrates are widely used in a variety of biological detections, the low sensitity and narrow dynamic range limit their applications in the assays that require rapid, robust and sensitive detection of biological molecules.  Fluorimetric detection is known to be 2-3 magnitudes more sensitive than colorimetric detection. However, there are very few fluorogenic peroxidase substrates available. 

We are pleased to introduce our serieal Amplite™ fluorogenic substrates to address this unmet need. Our Amplite™ ADHP is a sensitive fluorogenic peroxidase substrate that has much lower background than the material from other commercial vendors. ADHP generates highly fluorescent resorufin that has maximum absorption of 571 nm and maximum emission of 585 nm. Unlike other HRP substrates such as dihydrofluoresceins and dihydrorhodamines, the air-oxidation of ADHP is minimal. So far ADHP has been known as the most sensitive and stable fluorogenic probe for detecting HRP and H2O2. ADHP has been widely used to detect HRP in many immunoassays. On the other hand, ADHP can also be used to detect trace amount of H2O2. The ADHP-based H2Odetection is at least one order of magnitude more sensitive than the commonly used scopoletin assay for H2O2. Because H2O2is produced in many enzymatic redox reactions, ADHP can be used in coupled enzymatic reactions to detect the activity of many oxidases and/or related enzymes/substrates or cofactors such as glucose, acetylcholine and cholesterol, L-glutamate, amino acids, etc.

Although ADHP exhibits very high sensitivity for detecting peroxidate activity either in a fluorimetric or colorimetric mode, it requires higher pH to reach its maximum fluorescence. We are pleased to introduce our Amplite™ ADAP that has better water solubility and stronger fluorescence than ADHP. Addtionally, our Amplite™  is the only fluorogenic peroxidase substrate that generates fluorescent precipitates upon the peroxidase-catalyzed oxidation.


辣根过氧化酶HRP,由于其具有很好的稳定性和低的分子量是用于组织化学和酶联免疫检测中极其重要的酶。虽然有不少显色剂和发光剂的底物用于测定HRP,但是用于过氧化酶测定的荧光底物却很少。
ADHP (10-acetyl-3,7-dihydroxyphenoxazine) 
也称为Amplex Red Reagent,是目前最稳定和最敏感的HRP荧光底物。本试剂已将高纯度的ADHP溶解于适当的缓冲溶液中,可直接用于HRP酶反应,具有高灵敏度并且应用方便。实践证明,在ELISA实验中,使用ADHP作为显色底物,可检测到最低浓度达10-15 的标记物。
使用荧光酶标仪检测时请选择:激发波长Ex=530-560 nm;发射波长Em =585 nm
